Abstract Withania coagulans (Stocks) Dunal (family: Solanaceae) is critically endangered medicinal plant which needs immediate attention for in vivo and in vitro conservation. The authors reported alginate encapsulation of micro-cuttings and efficient in vitro plantlet regeneration in W. coagulans for rapid multiplication, short term storage and germplasm distribution. The in vitro established multiple shoot cultures were used as source of micro-cutting for encapsulation. Encapsulation of micro-cutting viz. shoot tips and nodal segments, with 3.0 % sodium alginate and 100 mM calcium chloride was found most suitable as it produced easy to handle beads with high (96.0 ± 4.0 %) regeneration capacity. Pulse-treatment of micro-cutting base with 2.46 mM indole-3-butyric acid for 2 min and subsequent encapsulation of these micro-cuttings produced the most suitable beads as these beads produced complete plantlets within 30 days. Attempts were made to utilize these beads for short-term storage and germplasm distribution. After storage at 4 °C under sterile conditions for 60 days, the entrapped micro-cuttings exhibited 72 % plantlet regeneration on 0.75 % agar-gelled Murashige and Skoog’s medium containing 1.11 µM 6-benzylaminopurine and 0.57 µM indole-3-acetic acid. The synseed-derived plantlets were successfully established and acclimatized. To the best of the knowledge, this is the first report on synthetic seed production in W. coagulans. Overexploitation and reproductive failure forced W. coagulans towards the verge of complete extinction and the encapsulation-regeneration system described here would facilitate genetic restoration programs, short term storage and germplasm distribution.
