Abstract The present investigation on “Molecular and phytochemical characterization of algal species from coastal belt of Okha region†was conducted at Department of Biotechnology, Junagadh Agricultural University, Junagadh with the objectives, phtyochemical characterization of different algal species from belt of Okha coast of Gujarat, to study molecular characterization of different algal species using PCR based molecular marker and to find out the phylogenic relationship among different algal species. To analyze molecular diversity of different algal species using PCR based molecular markers viz. Random Amplified Polymorphic DNA (RAPD), Inter Simple Sequence Repeats (ISSR). Total 20 RAPD primers were amplified to generate the 191 fragments in which 190 bands were polymorphic with 164 shared and 26 unique bands having an average of 9.5 bands per primer. The Polymorphism Information Content (PIC) values for RAPD marker were ranged from 0.69 (OPG-04) to 0.91 (OPP-05) with an average value of 0.85 per primer and RAPD primer index (RPI) differed from 2.45 (OPN-06) to 14.56 (OPP-05) with an average value of 7.9 per primer. Total 20 ISSR primers were used which generated 204 fragments in which all 204 bands were polymorphic having 183 shared and 21 unique bands with an average of 4.7 bands per primer and no monomorphic bands found. The Polymorphism Information Content (PIC) values for ISSR marker were ranged from 0.81 (UBC-810 and UBC-861) to 0.91 (UBC-847) with an average value of 0.85 per primer and ISSR primer index (IPI) differed from 5.69 (UBC-861) to 13.78 (UBC-847) with an average value of 8.85 per primer. The similarity coefficient of clusters analysis was ranged from 12.6% to 45.8% for RAPD. In ISSR marker, similarity coefficient was ranged from 17% to 62%. Solieria robusta and Sarconema filiforme species showed maximum similarity (62%). Spatoglossum asperum had maximum variability (24%) among other species. The collective data of RAPD and ISSR markers used to generate mixed clustering pattern to confirm the difference and similarity between species as the reliability of one particular marker does not fulfill the goal of identification of species and similarity coefficient was ranged from 24 to 62%. Dictyopteris delicatula and Udotea flabellum are the species exhibits statistically at par as well as higher mean value compared to others for total phenol, carbohydrate, flavanoid and chlorophyll that recorded 35% similarity and shared same cluster in RAPD as well as pooled data of RAPD and ISSR was concerned. The total protein and lipid was found to be the highest in Dictyopteris delicatula i.e., (8.5%) and (10.42%) respectively, while total carbohydrate in the same species is found to be the lowest i.e., (3.96%). The quantitative analysis of protein content ranged between 5.27% to 8.5% in Solieria robusta and Dictyopteris delicatula, respectively. Furthermore, Dictyopteris delicatula (10.42%) shown to have the highest percent of lipid content followed by Udotea flabellum (10.27%) and Caulerpa Lenthilifolia (10.04%). Finally, the fatty acid composition ranged as follows: 31–86.51% SFAs, 13.49–69% UnSFAs, 8.93–39.12% MUFAs, and 4.56–46.71% PUFAs. Therefore, from the present experiment, the data generated might be useful to some extent for the identification of the genetic diversity among 15 algal species which could be useful for algal strain improvement programs. However, the algal species fall in a same group showed wide variability in biochemical parameter except the species Dictyopteris delicatula and Udotea flabellum.
